PDRN — the ingredient everyone is talking about on TikTok. Hype or revolution?

PDRN (polydeoxyribonucleotides) is an ingredient with a solid medical history and serious clinical studies. The problem is that TikTok has mixed up the doctor-administered injection with a serum costing a few dozen zlotys from a chain store. Let’s break it down — without the hype, without the panic, with science.

“Salmon DNA” — sounds like a joke. But 2 billion views on TikTok don’t lie.

Go to TikTok and type in #PDRN. Or #SalmonDNA. Or #polynucleotides. You’ll see Korean dermatologists doing injections, influencers showing “glass skin after PDRN” and comments like “OMG need this NOW where do I get it.”

Then go to Allegro and you’ll find “salmon DNA serum” for 39 zlotys. On Temu for 12 zlotys. On Amazon for 25 dollars.

And here’s where the problem starts. Because PDRN is not a cosmetic buzzword to throw on a label. It’s a substance with a serious medical history, solid clinical studies — and very specific limitations regarding how it can work.

What is PDRN? (And how does it differ from PN?)

PDRN = polydeoxyribonucleotides. These are fragments of DNA — short chains of nucleotides, most often derived from salmon DNA (species Oncorhynchus mykiss or Oncorhynchus keta).

Salmon DNA has 97% homology with human DNA. So at the nucleotide sequence level, it’s almost identical. This makes the body “recognize” these fragments as its own and not react with rejection.

PN = polynucleotides. This is a broader term — it includes both PDRN and other fragments of nucleic acids (including RNA-derived). In clinical practice, PDRN and PN are used interchangeably, although PDRN is better studied.

How does it work? Biological mechanism

The mechanism is elegant and well documented:

1. Activation of adenosine receptor A2A — PDRN binds to this receptor on the surface of fibroblasts (cells that produce collagen). The activated receptor triggers the MAPK/ERK signaling cascade.

2. Proliferation of fibroblasts — more fibroblasts = more “factories” producing collagen, elastin, and hyaluronic acid. A study in Molecular Medicine Reports (2019) showed that PDRN can increase fibroblast proliferation by 30–40% in vitro.

3. Anti-inflammatory effect — PDRN inhibits TNF-alpha and IL-6 (pro-inflammatory cytokines). Less inflammation = better regeneration.

4. Stimulation of angiogenesis — the formation of new blood vessels. Better vascularization = better skin renewal.

Sounds good? Because it sounds good. And more importantly — it’s not marketing. It’s peer-reviewed science.

Where does this trend come from? Korea → TikTok → the world

  • 2010–2018: PDRN used medically in Korea and Italy mainly for wound healing, burn treatment, and tissue regeneration. In medicine — not in aesthetics.
  • 2019–2022: Korean dermatology clinics start using PDRN in facial mesotherapy. “Rejuran” (a Korean brand with PDRN) becomes one of the most popular treatments in Gangnam.
  • 2023–2024: TikTok picks it up. #SalmonDNA explodes. Influencers from Seoul show “before/after” — part of the effect is swelling from the injection, not a long-term result. 2+ billion views.
  • 2025–2026: PDRN enters the mainstream in Europe and the USA. Injectable products (Nucleofill, Plinest, Rejuran) available in aesthetic medicine offices. At the same time: an avalanche of “salmon DNA” cosmetics priced from a few dozen to several hundred zlotys.

And here we come to the crux of the matter.

PDRN serum vs injection in the office — they are NOT the same

Problem #1: Molecular size

PDRN is a polymer with a molecular weight of 50–1500 kDa (kilodaltons). The epidermis allows molecules with a mass of up to about 500 Da to pass through. That’s a difference of a thousand. The PDRN molecule is too large to penetrate the epidermis. Even with “penetration technology,” “liposomes,” or “nanocarriers” — there are no credible studies confirming that cosmetic serum with PDRN delivers the active substance to the dermis, where fibroblasts are located.

Problem #2: Concentration

Injectable products (Nucleofill, Plinest) contain PDRN in high concentration in a highly purified form. Cosmetic serum? The manufacturer does not have to disclose the concentration. And usually does not — because the concentration is symbolic.

Problem #3: Purity

Medical PDRN undergoes multi-step purification — proteins, lipids, and other contaminants from salmon DNA are removed. What remains is pure fragmented DNA. Cosmetic “salmon DNA extract”? It may contain a whole spectrum of substances — including proteins that can trigger an allergic reaction.

Verdict: Cosmetic serum with PDRN may hydrate the epidermis (thanks to other ingredients — glycerin, HA). But the effect of PDRN on fibroblasts in topical form is not confirmed by credible studies. An injection in the office is a completely different level — different penetration, different concentration, different effects.

PDRN vs hyaluronic acid vs collagen stimulators — what is what

Hyaluronic acid (HA)

What it does: Hydrates. Binds water (1 g HA = up to 6 liters of water). Fills (fillers). Mechanism: Physical — water + volume. Does not stimulate collagen production. Duration of effect: Immediate (filling). Fades after a few to several months. For whom: Volume loss, wrinkles, hydration.

Collagen stimulators (Sculptra, Radiesse, Profhilo)

What they do: Stimulate the production of your own collagen. Mechanism: Induce a controlled inflammatory response (Sculptra — poly-L-lactic acid) or provide scaffold molecules (Profhilo — slow-/fast-crosslinked HA). Duration of effect: Several weeks (collagen building). The effect can last one to two years. For whom: Loss of collagen, skin laxity, facial remodeling.

PDRN/PN (Nucleofill, Rejuran, Plinest)

What it does: Regenerates at the biological level. Activates fibroblasts, stimulates collagen, elastin, and HA. Has anti-inflammatory effects. Improves microcirculation. Mechanism: Biological — activation of A2A receptor, MAPK/ERK cascade. Duration of effect: 2–4 weeks (regeneration). Optimally after 3–4 sessions. For whom: Thin/tired skin, acne scars, regeneration after treatments, anti-aging prevention.

HA fills. Stimulators build collagen. PDRN regenerates the entire tissue. They are not substitutes — they are complements. The best protocols combine 2–3 of them.

Is PDRN safe?

Short answer: yes — in office conditions, in certified preparations.

  • PDRN has been used medically for over 20 years (mainly orthopedics, surgery, dermatology).
  • It comes from purified salmon DNA — all proteins are removed in the medical production process. No proteins = minimal risk of allergies.
  • Clinical studies on thousands of patients have not shown serious adverse effects.
  • The most common side effects of injections: swelling at the injection site (1–2 days), slight redness. Rarely: minor bruising.

PDRN is an invasive procedure (injection). It should be performed by a doctor or a qualified cosmetologist (depending on the depth of injection and national regulations). Not at home. Not by yourself. Not from a kit from Temu.

Skepticism where it’s needed

What we KNOW (solid evidence)

- PDRN can stimulate fibroblasts in vitro (laboratory studies) ✓ - PDRN often accelerates wound healing (clinical studies, meta-analyses) ✓ - PDRN in injection improves skin texture and hydration in many patients (clinical studies) ✓

What we DON’T KNOW (or know uncertainly)

- Whether the effects of PDRN on the skin are better than collagen stimulators (lack of head-to-head studies with Sculptra or Profhilo) - How long the effects last (long-term data limited — most studies up to 6 months) - Whether cosmetic serum with PDRN has any biological effect (lack of credible studies)

  • “Salmon DNA serum” for a few dozen zlotys — probably worthless in terms of PDRN. It may hydrate (other ingredients), but PDRN will not reach fibroblasts.
  • Offices offering “PDRN” without the name of the preparation — ask for a specific brand (Nucleofill? Plinest? Rejuran?). Noname products may have questionable quality.
  • Promises of “effect after 1 session” — real effects of PDRN build up over 3–4 sessions. If something looks dramatic after 1 treatment — it’s often swelling, not regeneration.

What you can do at home (realistically)

If you’re not planning a treatment yet — here are ingredients with proven effects that you can incorporate into your home care:

  • Serum with 5% niacinamide + peptides — not PDRN, but proven stimulation of collagen at the epidermal level.
  • Retinal (retinaldehyde) — the gold standard of anti-aging with the longest evidence base. Stimulates cell renewal, collagen, elastin.
  • SPF 50 daily — 80% of skin aging is photoaging. The best “anti-aging” is UV protection.
  • Collagen supplementation (hydrolyzed, minimum 3 months) — studies suggest improvement in skin elasticity and hydration.
  • 7–8 hours of sleep — fibroblasts are most active at night (peak regenerative activity: 11:00 PM–2:00 AM). Lack of sleep = lack of regeneration, no matter how much serum you apply.

And if you really want PDRN?

Then go to the office. Not to TikTok. Not to Allegro.

PDRN/PN injection is a medical or para-medical procedure. It requires consultation (skin assessment, indications, contraindications), sterile conditions, certified preparation (Nucleofill, Plinest, Rejuran — registered as medical devices) and should be performed by a qualified person.

The standard protocol is 3–4 sessions every 2–4 weeks, then maintenance 1–2 times a year. The effects build up gradually — the skin becomes denser, better hydrated, more elastic.
