Retinol, peptides, hyaluronic acid — what do they really do in anti-aging?

Retinol, peptides, hyaluronic acid. Three names that appear on almost every anti-aging cosmetic package. Together they sound like a complete answer to aging — and indeed each of them has its place in this story. But if someone has ever tried to understand them, they quickly hit a wall of marketing slogans. What does it mean that something “stimulates collagen”? Or that it “hydrates deep within the skin”? And why does a retinol product have to be used in the evening, while a hyaluronic acid serum can be applied at any time? These questions have concrete answers — biological, not advertising. In this text, we break down each of these ingredients into their basic components: how they reach the skin, what they do there, and what they can realistically change. Without the noise, layer by layer.

Three ingredients — three completely different mechanisms

Before we address each of them individually, it’s worth understanding one key fact: retinol, peptides, and hyaluronic acid are not three versions of the same action. They are three different languages that cosmetics speak to the skin. Each works at a different level, through a different mechanism, with a different time horizon for effects. Therefore, putting them on the same shelf as “anti-aging ingredients” is a bit like comparing architecture, painting, and heating as “renovation work” — each is needed, but none can replace the others.

  • Retinol — speaks to skin cells in their own language, mimicking natural signals that regulate growth and differentiation. It works at the level of cell nuclei.
  • Peptides — are short chains of amino acids that act like biological messages: they tell the skin what to produce or what to stop. Different peptides send different messages.
  • Hyaluronic acid — does not send any commands. It acts like a sponge: attracting and retaining water, giving the skin volume and elasticity here and now.

A good metaphor is a factory. Retinol changes production management — it issues directives deeper in the skin. Peptides are orders from customers — each peptide tells the factory what is currently missing. Hyaluronic acid is not a message to the factory, but a ready raw material — we bring it from the outside because the factory produces less and less of it with age.

Retinol: the most researched anti-aging ingredient

Retinol is a derivative of vitamin A and one of the best-documented cosmetic ingredients overall. Its advantage over most “revolutionary novelties” is simple: it has been working for decades, and research confirms this. But to understand how this is possible, one must go down to the level of cells.

After being applied to the skin, retinol penetrates through the epidermis and is converted in the dermis — first into retinal, and then into retinoic acid (tretinoin). It is retinoic acid that is the active form. It binds to receptors inside cells that act as genetic switches — literally influencing which genes turn on or off. In practice, this means, among other things, accelerating the renewal of epidermal cells, limiting the activity of enzymes that break down collagen, and stimulating fibroblasts — cells in the dermis responsible for producing collagen and elastin.

Irritation, dryness, and peeling at the beginning of retinol use are not a mistake — they are a sign that the ingredient is working. Accelerated turnover of epidermal cells causes old cells to shed faster before the skin has time to adjust. In many people, the skin adapts within a few to several weeks. That’s why experts recommend starting with lower concentrations and using retinol gradually — not out of whim, but with the thought that the skin keeps up with the changes.

Retinol used in cosmetics is at a lower concentration than the prescription tretinoin — which is why it works more gently and slowly, but with regular use, it gives visible effects for many people. Retinol is photolabile (it breaks down under light) and can increase the skin's sensitivity to the sun — hence the recommendation for evening use and mandatory SPF filter in the morning.

What retinol can and cannot reverse

Can: accelerate epidermal renewal, support collagen and elastin production, influence sunspots, smooth texture. Cannot reverse: loss of facial bone, changes in the geometry of the oval, deep gravitational grooves — these processes occur much deeper than the reach of cosmetics. Therefore, retinol is an important element of care, but one of the elements — not the entire system.

Peptides: biological messages to the skin

Peptides are short chains of amino acids — the building blocks of proteins. The skin itself is full of peptides: it uses them as internal signals regulating repair, production, and degradation of tissues. Cosmetic peptides try to mimic or enhance these signals from the outside.

The mechanism is elegant in its simplicity. When collagen degrades (e.g., due to time, sun, or enzyme activity), its fragments — called matrikines — travel in the skin as an alarm signal: “collagen is lacking, we need to produce more.” Synthetic peptides can mimic these fragments and send the same signal to fibroblasts, not waiting for actual degradation. It’s a bit like ordering replenishments before the warehouse runs out.

  • Signaling peptides — stimulate fibroblasts to produce collagen, elastin, or other components of the skin matrix.
  • Carrier peptides — transport minerals (e.g., copper) to places where they are needed for collagen synthesis.
  • Inhibitory peptides — block neuro-muscular signals responsible for the tension of mimic muscles (the so-called “botox in cream” — a marketing analogy, the mechanism does exist, although it works much weaker).
  • Barrier peptides — support the reconstruction and integrity of the epidermal barrier.

A popular question: “if I need collagen, why not apply it directly?” The problem is structural: the collagen molecule is too large to penetrate the epidermis. It’s like trying to smuggle furniture through a closed window. Peptides are smaller — they can penetrate deeper and instruct the skin to produce collagen from within, rather than delivering it from the outside. It’s the difference between sending building materials and sending an architect.

Peptides are generally well tolerated by the skin and rarely cause irritation — which is why they are often used in formulations for sensitive skin or in combination with retinol (as a soothing ingredient). The effects are subtler and require regularity: with occasional use, it’s hard to expect visible change.

Hyaluronic acid: hydration as a structural function

Hyaluronic acid (HA) is a substance naturally present in the human body — in the skin, joints, and eyeballs. In the skin, it plays a fundamental role: binding water, giving the tissue elasticity, volume, and smoothness. Fibroblasts produce it throughout life, but with age, production gradually decreases — hence the skin becomes thinner, flatter, and loses its characteristic “fullness.”

But not every hyaluronic acid works the same — and this is the crux that cosmetics often do not explain. The key parameter is molecule size. Large HA molecules remain on the surface of the skin and create a moisturizing film — providing an immediate effect, visible almost right away, but superficial. Small molecules can penetrate deeper, into the upper layers of the epidermis, where they store water within the tissue. The latest formulas use fragments of HA of different sizes simultaneously — to act on several levels at once.

Many people confuse cosmetic hyaluronic acid with that used in aesthetic medicine offices. This is an important distinction: injection fillers are chemically cross-linked HA — creating a gel that maintains shape and volume in the tissue for many months. Cosmetic HA is not cross-linked, is not injected, and does not serve as a filler. It works completely differently — as a moisturizing ingredient. These are two different technologies, although the same molecule.

Cosmetic hyaluronic acid is a safe and well-tolerated ingredient, even for sensitive skin. One practical note: in dry air (low humidity, air conditioning), large HA molecules can paradoxically pull water from deeper layers of the skin to the surface, and then to the environment — which in some people gives a feeling of dryness. Therefore, after a serum with HA, it’s worth sealing it with a cream that limits evaporation.

How these three ingredients talk to each other

When we understand the mechanism of each ingredient individually, a practical question arises: does it make sense to use them together? And does the order of application matter? The answer to both is: yes — and here is a simple reasoning.

Important caveat: for many people, there is no need to use all three at once. Sensitive skin may react to retinol with irritation — then it’s worth starting with just peptides and HA, and introducing retinol gradually. The goal is not to overload the skin with “the best ingredients,” but to tailor them to the specific needs of a particular complexion.

Which ingredient might your skin be lacking?

The boundary between cream and clinic

The conversation about retinol, peptides, and hyaluronic acid inevitably leads to the question that many ask in silence: when does cream stop being sufficient? There is no magic boundary here — but there is logic.

Cosmetics work on the epidermis and upper layers of the dermis. They can improve hydration, texture, complexion, slow down certain processes. This is real and valuable. But some changes occur deeper: in the collagen scaffold, in adipose tissue, in the oval of the face. No cream physically reaches there — and no manufacturer, to be honest, can promise otherwise. The boundary does not lie between “good” and “bad” products, but between the depth of problems and the reach of the tool.

There is a big difference between skin that lacks hydration and good care, and skin that needs deeper work than cosmetics. The first will respond to wisely chosen retinol and peptide serum — and the change can be really noticeable. The latter will find a good cream to be a sufficient complement, but not an answer to the core issue.
